Abstract

We demonstrated light sheet fluorescence imaging at sub-micrometer resolution of biological samples enclosed in PDMS soft lithography devices. This was achieved by minimizing the optical distortion induced at the external PDMS device interface using a sucrose solution with refractive index (RI) matched to the PDMS. A commercially available inverted Selective Plane Illumination Microscopy (iSPIM) was modified with a single mode fiber (SMF) and an electrically tunable lens (ETL) module to facilitate RI matching and compensate for sucrose solution induced axial focal shift. We termed this protocol as the modified iSPIM (m-iSPIM).
