This was achieved by minimizing the optical distortion induced at the external PDMS device interface using a sucrose solution with refractive index (RI) matched to the PDMS. A commercially available inverted Selective Plane Illumination Microscopy (iSPIM) was modified with a single mode fiber (SMF) and an electrically tunable lens (ETL) module to facilitate RI matching and compensate for sucrose solution induced axial focal shift.
